A bit unusual -
A senior manager will send 4 ladies to DC for a conference, but they must share two rooms due to budget restrictions.
One of the 4 ladies is a lesbian and none of the other three are comfortable sharing a room with her.
They have privately expressed this discomfort with the senior manager even though one of the initially said she would share.
The rooms are booked, the flights are paid for and except for this last minute glitch, it's ready to go.
How would you handle this?

How do you know the lady is a lesbian? Has she come out?
If yes then the Senior Manager needs to change his decision or not make the conference mandatory.

Yes, the lady is out.
The conference is not mandatory, just the budget driven need to only pay for two rooms.

I would tell them they are working out the particulars of who is rooming with who on their own. The orientation of the one employee is irrelevant. To treat her differently, e.g give her a private room while the other 3 share is as inappropriate as allowing the 3 to object to sharing a room with her simply because she is a lesbian.
I wouldn't let myself get dragged into this if at all possible. The rule is 2 rooms for 4 same gender women. They can choose their roommates. The company will not reimburse for private rooms and will only pay for these 2 rooms.

Hi:
HRPro's post is totally on point. Also, some states protect employees on the basis of sexual orientation, which a taffy pull regarding this matter raises various legal issues.

As a collateral point, a few organizations (e.g., Walmart) mandate sharing of hotel rooms--so this is not an uncommon practice.

I agree with the PP. Do these women think just because she is a lesbian that she is going to assualt them or something? If they are worried about nudity and that sort of thing, the best bet would be to just change in the bathroom. I don't know about others but I sure wouldn't want to change in front of any hetrosexual co-workers nor would I want them to change in front of me. That would be too uncomfortable.

Well these women must think they're something special, because if they have to share a room with a lesbian obviously she would be after them! Are you kidding me?? If these women have worked with her and she's not shown any interest in them before, nothing will change just because they share a hotel room for business. Lesbians are professionals, too, just like the other ladies. They don't use business trips for personal pleasure purposes. Grow up ladies.
