two of our managers recently quit leaving a staff of 4 managers in charge of a 365 days/24 hour call center with 465 agents and 38 supervisors.
The GM told those 4 managers they need to pick between

I agree that "Proving oneself in times of pressure ultimately reaps reward." But they shouldn't expect the rewards to come from the type of cement-headed Neanderthol management the OP describes.
If this kind of ultimatum works once, they will do it again and again. This is what happens when you cut down a forest without replacing the trees.

Even if they don't do it again, it will be remembered.
Honestly I would try to brainstorm WITH THEM a better solution. For example, I am not sure why their two days off can't be consecutive? I am betting that they can come up with solutions that you may not have thought of.
And I am also not sure why they can't hire a temporary position for x months.
It does sound like they are going to be punished because someone else quit and there wasn't any backup or succession planned. Either way, you are risking bad blood with all 4 that are left.
