Company has been doing a lot of hiring some external and internal. An employee applied for a management position, informed their immediate supervisor they were applying. the supervisor advised the employee that it would not be wise for them to apply for the position. Due to they are not a people person and they do not know a lot of the staff in the department. Whether the comment was true or not.Is this appropriate for a supervisor to discourage an employee from applying for a position.

There is no apparent legal issue here unless the discouragement was based on a protected characteristic such as race, gender, national origin, religion, etc

Appropriate is a very subjective term. To me the information provided to the applicant, while probably accurate, is not appropriate.

First the applicant should be praised for:
1. Advising the supervisor and keeping the line of communication open
2. Aspiring to improve themselves.

It could then have been used as a "teaching moment" and they could have had a chat regarding the job requirements both on paper and off the JD.

Employees need feedback but there is a bit of art in how to deliver it and make it lemonade from lemons.

in the hard times companies import some of the assests in internal  external matters for the  good policies.

It's hard to say without knowing the actual dynamics of the company and the individuals.  In some cases, I can see where it would be helpful and could be used as a teaching moment.  In other cases, if the supervisor has the authority to block the change/transfer, I can see where it is best to be upfront about it in the beginning.

I could also see it having negative impact if for example the supervisor also applied for that position and is discouraging the other employee because he himself wants the position.

Inappropriate? Depends....Against company standards/policies? Possibly...But there is nothing in what you have written that is illegal.

I would say it is important for a manager to have a discussion with the employee to better understand their motivation to move, but I would not say it is appropriate to tell someone "they are not a people person."  Coaching is about drawing out good qualities in an individual, and developing as appropriate rather than shutting someone down.
