our dept is about to experience a reduction in staff through attrition. I anticipate being asked to take on more of my departing colleague's repsonsibilities, who will likely not be replaced. Would it be out of line to negotiate an increase in pay to go along with the additional workload?

Depends. If your departing colleague isn't being replaced because your company can't afford to pay a replacement, then no, this wouldn't be a good time to be asking for more $.
